Description

This Mouse Semaphorin 4G ELISA Kit is intended for quantitative detection of mouse SEMA4G in cell culture supernates, serum and plasma (heparin, EDTA, citrate). Strip well format. Reagents for up to 96 tests.
This mouse SEMA4G ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for SEMA4G has been precoated onto 96-well plates. Standards (Expression system for standard: NSO, Immunogen sequence: V18-M673) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for SEMA4G is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse SEMA4G amount of sample captured in plate.
The capture antibody is a monoclonal antibody from rat, the detection antibody is a biotinylated polyclonal antibody from goat. Expression system for standard: Semaphorin-4G is a protein that in humans is encoded by the SEMA4G gene. This gene is mapped to 19; 19 C3. Semaphorins are a large family of conserved secreted and membrane associated proteins which possess a semaphoring (Sema) domain and a PSI domain (found in plexins, semaphorins and integrins) in the N-terminal extracellular portion. Based on sequence and structural similarities, semaphorins are put into eight classes: invertebrates contain classes 1 and 2, viruses have class V, and vertebrates contain classes 3-7. Semaphorins serve as axon guidance ligands via multimeric receptor complexes, some (if not all) containing plexin proteins. This gene encodes a class 4 semaphorin. This gene and the gene for mitochondrial ribosomal protein L43 overlap at map location 10q24.31 and are transcribed in opposite directions
